PROTAC SGK3 degrader-1 (SGK3-PROTAC1), is a potent SKG3 degrader based on von Hippel-Lindau ligand. PROTAC SGK3 degrader-1 (0.3 μM) induces 50% degradation of endogenous SGK3 within 2 hours, with maximal 80% degradation observed within 8 hours, accompanied by a loss of phosphorylation of NDRG1 (an SGK3 substrate).
体外研究：
PROTAC SGK3 degrader-1 (SGK3-PROTAC1) (0.3 μM, 4 weeks) 联合 GDC0941 抑制 CAMA-1 或 ZR-75-1 细胞的生长。PROTAC SGK3 degrader-1 (0.1 μM, 48 h) 可使 HEK293 细胞中 SGK3 水平降低 65%，而不影响SGK1、SGK2 和 S6K1 的表达水平。PROTAC SGK3 degrader-1 (1-10 μM, 48 h) 适度降低 HEK293 细胞中 S6K1 水平。PROTAC SGK3 degrader-1 ( >0.1 μM, 8 h) 可降低 HEK293 细胞中 NDRG1 的磷酸化，诱导 CAMA-1 和 ZR-75-1 细胞中 SGK3 的降解，但不诱导 SGK1 或 S6K 的降解。
